Sequential designs for dose escalation studies in oncology. (English) Zbl 1347.62232

Summary: In Phase I dose escalation studies the 3 + 3 design is a commonly used method. We suggest a modification of this method to include a parametric model and the concepts of optimal design theory, with the aim of improving this method. The performance of the original and the new approach are investigated by simulations.
